Western blotting: Quantify with confidence

Support and solutions for clear, quantitative protein detection to accelerate drug research and discovery
7 steps to western blotting

Learn how to get more from your Western blots

In research, you know reliable protein analysis is key to identifying molecular targets, quantifying expression, and uncovering tomorrow’s big breakthroughs.

Along with a complete workflow, we offer Western blotting training, education, and troubleshooting tools to help you avoid common pitfalls, minimize variability, and deliver data you can trust.

Reproducible results with every blot

The first time and every time, get consistent Western blot performance with a standardized, scalable workflow.

  • Reagents: enhanced chemiluminescence (ECL) and standards for sensitive, low‑background detection delivering a clear, lasting signal
  • Membranes: nitrocellulose (NC) and polyvinylidene fluoride (PVDF) options offering high protein binding capacity and retention
  • Imaging systems: quantitative, low-background imaging optimized for high signal-to-noise ratios
  • Software: chromatogram visualization to annotate and compare bands to peaks, with an image integrity checker enabling faster publication

Built on decades of Western blotting expertise

From pioneering ECL detection to supporting today's best practices in protein analysis, we help researchers achieve reliable, reproducible results.

Sensitive detection for low-abundance proteins

Our ECL reagents support diverse applications, offering sensitive detection across a wide dynamic range so you can accurately measure low-abundance proteins and compare expression across samples.

Accurate and robust protein quantitation

Total protein normalization is becoming the preferred method for protein quantitation, offering a broad detection range with reduced susceptibility to biological variation. Learn more in our quick explainer video.

7 steps to Western blot workflow optimization

Step 1 Sample preparation and protein extraction

Enhance protein stability, reduce background noise, and improve resolution through careful sample preparation.

Step 2: SDS-PAGE protein separation

Select the appropriate gel percentage and use a stacking gel to achieve tight, well-resolved protein bands during electrophoresis.

Step 3: Protein transfer to membrane

Choose between wet or semi-dry transfer methods and between PVDF or nitrocellulose membranes, but keep in mind that other factors like buffer composition and electric field strength also influence blotting success.

Step 4: Membrane blocking

Block to prevent nonspecific binding of primary and secondary antibodies.

Step 5: Primary antibody incubation

Optimize the concentration and incubation time for the primary antibody.

Step 6: Secondary antibody incubation

Select a secondary antibody conjugated to either a chemiluminescent or fluorescent detection molecule. Stripping and reprobing your membrane enables reuse of the blot to detect additional proteins.

Step 7: Detection and analysis

Western blotting imaging options include x-ray film, CCD cameras, or laser scanners, depending on the detection method used.

Trust in Western blot data starts with image integrity

You need to know that your raw image data is authentic and intact.

Our imaging systems, analysis software, and image integrity checker software help you verify image authenticity, confirm which files are original, and support paper submission with a report you can attach.

Alongside the software, we provide educational resources that help you strengthen image handling practices and support scientific integrity.
