We recommend using PuReTaq for amplicons up to 3 kb.
In our laboratories PuReTaq is routinely used in real- time PCR applications using both fluorescence resonance energy transfer (FRET) and intercalating dyes.
The concentration of Mg2+ is kept low at 1.5 mM. Users may add more Mg2+ to optimize the signal-to-background ratio and improve the lower limits of detection and reproducibility of sequence detection at low copy numbers. Addition of Mg2+ may also improve the binding strength of the oligonucleotides used for priming.
The final concentration of KCl is 50 mM
PureTaq is formulated with a recombinant wild-type Taq polymerase that lacks proofreading activity. Therefore, error rates are relatively high at > 1 × 10-5.
We achieve sensitivities below 10 copies of lambda DNA in qPCR.
