Product documentation—a user manual, including a certificate of conformance, is provided. The certificate of conformance shows conformance with 21CFR177, USP Class VI, and animal-free origin (or EMEA/410/01) for each wetted part material as well as traceability of the material. The product documentation contains a certificate of analysis showing packing performance as well as endotoxin and microbiology test results for the delivered column. The user manual is distributed with each column and the CoA can be downloaded from gelifesciences.com/certificates.
Validation guides—gives access to product information of the ReadyToProcess column including stability, validation, and quality as well as a brief description of the production process.

Use the sample supplied with the column. Check the label for Lot No.
Rigorous transport studies have shown that there is no change in column performance after shipping. Beta testers confirm splendid performance. Shipping and drop tests are included in Regulatory Support File.
Possible causes |

Column is clogged. |
1. Perform regeneration of the column |
Bent tubing. |
Check that the flow path is not restricted. |
Buffer viscosity too high. |
Check the viscosity of all buffers. Viscosity is a function of temperature. (Lower temperature gives higher viscosity.) Let low-temperature buffer reach operating temperature before starting the run. |
Microbial growth in buffers. The buffer normally become opalescent due to microbial growth. |
Check buffers, especially those with phosphate, for microbial growth. Replace with fresh buffer if necessary. |
Sample and collection vessels at different levels. |
Adjust the vessels to approximately the same level. |
The prefilter might be blocked. |
Check the prefilter. |

Accessories
1. Assemble the Normal flow filter (NFF) inlet to the flow kit.
2. Evacuate air by filling NFF with liquid. Use the system to pump liquid through the system to the NFF.
3. Assemble the NFF to the column inlet. Extra tubing may be needed to prolong the flow kit. Please make sure the tubing is not folded and does not contain any air.
