When running FlexStand Holder with peristaltic pump, the maximum pressure that can be achieved is 2 bar g (30 psi g).

The use of a permeate flow control is strongly recommended during upstream clarification of target material. 

A permeate flow control kit together with a pump for the flow rate is required for permeate flow control. Permeate flow control rate is dependent on the process flux (LMH) and surface area. 

Detailed information about Permeate control valve (56-4105-89) and Permeate flow control kit (56-4107-73), is available below.

Suitable peristaltic pump (56-4106-53) for permeate flow control, flow rates 5.4 ml/min - 2 liter/min. Information available below.

Countries using 220 V power, have to order power cords separately for the pump above and system.

Feed pump (11-0005-66 or 11-0005-67), Feed reservoir (56-4107-67 or 56-4107-70), Outlet pressure gauge for FS-03LVS (56-4106-04), Outlet pressure gauge for FS-03LVS-30 ( 56-4106-03) and Tubing (56-4106-19, 56-4106-22 or 56-4105-23) have to be ordered separately.

After use, store the cartridge with liquid inside using blank off cap, gasket and clamp on the feed and retentate ports.

Seal the permeate port with a short piece of tubing i.d. 9.5 mm, which is secured using snapper clamp or cable tie wrap. The end is closed with snapper clamp or cable tie wrap.

Issue: This technical brief will describe the fittings and tubing required to physically connect a size 3M(A) or 4M(A) hollow fiber cartridge to the ÄKTAcrossflow system. The maximum feed flow rate on ÄKTAcrossflow is 600 ml/min.

Connection of Size 3 Hollow Fiber Cartridge to ÄKTAcrossflow:

 

Feed & Retentate Ports: Place gasket between end port and connector 25 mm TC - 5/16 female, (18-1169-22). Secure with clamp.

Lower Permeate Port: Connect tubing i.d. 3.1 mm or 6.4 mm (56-4106-17, PTSL03-10 or 56-4106-18, PTPM06-10). Fold over tubing. Secure with cable ties.

Upper Permeate Port: Connect at least 2.5 cm tubing i.d. 3.1 mm or 6.4 m. Secure with cable ties.

Connect connector Luer female - 3.1 mm (0.125 inch) barb (56-4105-75, RBFL-1) >> M6 female - Luer male (11-0026-43) >> 5/16 female - M6 male (18-1127-76) >> system permeate connection.

 

Connection of Size 4 Hollow Fiber to ÄKTAcrossflow:

 

Feed & Retentate Ports: Place gasket between end port and connector 25 mm TC - 5/16 female, (18-1169-22). Secure with clamp.

Lower Permeate Port: Connect tubing i.d. 6.4 mm (56-4106-18, PTPM06-10). Fold over tubing. Secure with cable ties.

Upper Permeate Port: Connect at least 2.5 cm tubing i.d. 6.4 mm. Secure with cable ties.

Connect connector Luer female - 3.1 mm (0.125 inch) barb (56-4105-75, RBFL-1) >> M6 female - Luer male (11-0026-43) >> 5/16 female - M6 male (18-1127-76) >> system permeate connection.

NOTE: Snapper clamps are delivered with hollow fiber cartridges. They may be used to secure tubing to the lower and upper permeate port barbs. Use pliers to secure the fitting. Check for leaks before loading sample.

General Advice for Troubleshooting when Using Cross Flow Filters

Please check the following items during troubleshooting:

Samples volume and system working volume have been used in calculations of concentration factor and diafiltration beforehand.

Process settings of Membrane Capacity, Flux, Shear (or volumetric cross flow rate) have been properly applied for both sample characteristics and membrane pore size or NMWC designation.

Filter is properly wetted before use (also make sure that there is no air lock in the filters created by condensation on microfiltration membranes as a result of incorrect autoclave settings).

Ensure CFF filters are properly wetted before starting processing. This may be tested against new cartridge “Clean Water Flux Test or against the water flux specifications in the Certificate of Quality. Microfitlration membranes are shipped dry and are rewetted with water or buffer. Hollow fiber ultrafiltration membranes are shipped in glycerol wetting preservative. These filters may require 15-20% alcohol/water solution to restore wetting or remove glycerol. Flat sheet cassette ultrafiltration devices (for example, KvickLab and KvickFlow Cassettes) are shipped in a solution of NaOH/Glycerol which must be flushed out before use.

Prior to using cassettes, make sure the single gasket is in the correct position in the holding device and that the cassette(s) within are oriented properly in the holder.

Check integrity of cassette system using water under pressure on retentate side to 1 barg to check for leaks in the seal of the cassette with the stainless steel holding device.

Check to see that cartridges or cassettes that have been previously used are checked for integrity (air diffusion or bubble point tests) and for cleanliness (normalized Clean Water Permeability before processing).
