Library preparation

Library preparation in genomics is the generation of DNA fragments for sequencing.

Library preparation for NGS involves creating a collection of DNA fragments for sequencing. Genomic libraries for next-generation sequencing (NGS) can be prepared through fragmentation of a genomic DNA sample and ligating adapters to both ends of the DNA fragment.

The purpose of a DNA library is to create a collection of DNA fragments spanning a gene of interest, a region of the genome or the entire genome for a sequencing instrument to read the genetic codes (i.e. DNA sequence). Reading the DNA sequences of the library helps us to understand the genome under study.
