Cryo electron microscopy (cryo-EM) is quickly becoming the preferred method for determining the structure of more challenging proteins. As a result, the need for micropurification has increased significantly. To address this challenge, Cytiva has developed the ÄKTA™ pure micro system and fraction collector F9-T for ÄKTA™ pure 25 protein purification system.
Sara Corin, General Manager, Downstream Hardware says: “Our new system was developed in response to the challenges our Structural Biology customers were experiencing. There was an urgent need to generate pure and homogenous samples in high concentration during cryo-EM studies. Often the sample volume was limited. With the launch of the ÄKTA pure micro, we can now provide our customers a better solution within this application area.”
Prior to cryo-EM, membrane proteins and larger protein complexes were too difficult to crystallize or too large to study with Nuclear Magnetic Resonance (NMR). Challenges when purifying proteins for cryo-EM experiments include limited sample volume in microliter (µL) scale and the inability of the chromatography system to handle limited sample volumes.
ÄKTA™ pure micro offers a flow path with low hold-up volumes and provides a comprehensive solution for small sample volumes and micro preparative columns. The system is equipped with 0.6 mL mixer, injection valve, 2 mm UV flow cell, conductivity monitor, and outlet valve. Together with appropriate tubing and connectors the system volumes are minimized, and high peak resolution is maintained throughout the flow path.
Scientists can also use the Micro kit to convert existing ÄKTA™ pure 25 M flow path for well optimized microscale purification. The Micro kit includes the same components and tubing as used in ÄKTA™ pure micro.
