High IgG tolerance
Downstream samples generally have high concentrations of IgG, which creates a challenge for protein A measurement. Accurate measurement requires dissociation of the ligand and IgG, which is especially tricky when the concentration of protein A ligand is very low (< 1 ppm) due to the large excess of IgG.
To determine the effect of IgG on dissociation, we spiked high, medium, and low concentrations of PrismA ligand into decreasing concentrations of IgG. The PrismA ELISA kit could accurately determine PrismA concentration in samples containing as high as 20 mg/mL of IgG

The PrismA ELISA kit is capable of detecting low amounts of ligand including at the final stages of purification. Leached protein A concentration in downstream materials is usually very low, so a highly sensitive assay is required to measure leached ligand in these types of samples.
The PrismA ELISA kit has a lower limit of detection (LLD) of 10 pg/mL and a lower limit of quantification (LLQ) of 10 pg/mL.

Confidence in results
It is good practice to demonstrate clearance of leached PrismA at each process step.
The PrismA ELISA kit has good compatibility with each process step, as seen by the parallel response in the calculated stock concentration and accurate recovery in the spiked control samples.

It is important to have an assay that can give accurate and reproducible results over multiple experiments. Assay precision is the coefficient of variation (CV) within a single assay (intra-assay) and across several experiments (inter-assay).
Intra-assay variation was assessed from the interpolated concentration of 127 in-process sample measurements.
96% of samples had in intra-assay CV < 10%, with a mean CV of 2.88%.
Inter-assay variation was assessed from 51 samples measured in triplicate assays. 78% of samples had an inter-assay CV of < 10%, with a mean CV of 5.95%.
A robust assay with excellent buffer compatibility
A robust assay should be compatible with all downstream buffers used in a particular process.
The PrismA ELISA kit was tested with common buffers used in downstream purification of monoclonal antibodies. The results showed the kit was compatible with all five buffers tested, as indicated by all dilutions of each sample (8/8) being within the acceptance criteria of the assay, and the parallelism being below the target of 20%.
