Adapter ligation

Adapter ligation is method of attaching synthetic oligonucleotides with known sequences to both or either ends of complementary DNA (cDNA) fragments. Due the unique adapter sequence, they can be used as (1) a barcode for multiplexing different samples, (2) a priming site for amplification or sequencing, (3) a hybridization sequence to bridge two DNA fragments.

This step can be checked by amplifying and then purifying the ligated fragments using size selection (e.g. solid phase reversible immobilization method with Sera-Mag Select or gel purification method) and quantifying on a BioAnalyzer instrument.
