Gentle tissue disaggregation for optimized cell viability and yield in single-cell omics research

By Cytiva

Single-cell omics enables the identification of sub-populations of cells that exist in a sample and the separate data signatures for each sub-population that indicate its size, expression and genome structure. Recent technological advancements have made single-cell omics (genomics, proteomics, metabolomics) an increasingly powerful tool for diagnosis, treatment response prediction, treatment selection and monitoring, especially for cancer. Researchers can analyze tens of thousands of individual cells from a single sample, gaining specific insights about cell populations and their behavior in diseased tissue. The initial sample disaggregation into a single cell suspension, however, is an unestablished part of the process. Current techniques commonly involve manual mechanical or enzymatic disruption. These techniques can result in losses via ruptured or attached cells, making them unsuitable for library preparation.

The VIA Extractor™* tissue disaggregator is designed for the disaggregation of human and animal solid tissue and tumor samples into viable, single cells. It uses a mild processing approach for consistently high cell viability, yield and preservation of cell integrity relative to the parent sample. The system provides a semi-automated process for use in high-throughput omics research, giving reliable results in single-cell sequencing and flow cytometry applications.

For omics applications, the VIA Extractor™ tissue disaggregator offers the following key benefits:

  • Gentle: Optimized cell viability and yield. Cells from the digestion process are unaltered and alive for later use in omics and single-cell sequencing.
  • Standardized: The system produces consistent output and yield, resulting in reduced sample to sample variation.
  • Semi-automated: Process is as simple as possible, reducing components and steps for tissue dissociation.
  • Time-saving: Solid tissue to single cell suspension in as little as 10 minutes.

To benefit from the full functionality of the system for your omics and single-cell research, the following components are available:

  • VIA Extractor™ tissue disaggregator: Device for disaggregation of human and animal solid tissue and tumors into single cells, used in combination with the VIA Freeze™ Uno controlled-rate freezer.
  • VIA Freeze™ Uno controlled-rate freezer: Liquid nitrogen-free controlled-rate freezer combining customizable freezing profiles, electronic control systems, and a conduction cooling method to optimize GMP-compliant cryopreservation.
  • Omics pouch: 3-compartment sample bag for digesting up to 1.2 g of tissue using enzymatic and mechanical disaggregation.
  • Omics clamp: Works with Omics pouch providing a secure seal after the sample insertion, while providing the support to the bag during tissue dissociation in the VIA Extractor™ tissue disaggregator.
  • Omics applicator: Syringe mechanism to aid insertion of tissue sample into the Omics pouch for enzymatic and mechanical tissue dissociation using VIA Extractor™ tissue disaggregator.
