Overview
Deaza dGTP is a nucleotide analog that is a candidate substrate for processive telomerase. Its use can improve Polymerase Chain Reaction (PCR) yields and makes sequencing of GC rich sequences easier.
- For reducing band compressions* in sequencing gels.
- Supplied as convenient solutions at pH 7.0 and 7.5. c⁷dGTP is supplied in purified water as 5 mM and 10 mM solutions.
Deaza dGTP is a substrate for most DNA polymerases, including Taq DNA polymerase. It can also be applied to dideoxy-chain termination sequencing methods, in place of dGTP to overcome compression problems in gel electrophoresis when sequencing GC-rich stretches of DNA. Partial substitution of Deaza dGTP for dGTP in PCR can improve the product yield when the template is GC-rich.
Technical Information:
*Band compressions are ambiguous band patterns on DNA sequencing gels. This is caused by stable, intra-strand secondary structures which are not fully denatured during electrophoresis. The likelihood of occurrence can be reduced through substitution of nucleotide analogs within sequencing mixers. To resolve G-band compressions, use 7-deaza dGTP (c⁷dGTP).
