Troubleshooting for Wave Bioreactor System 200 and Wave Bioreactor System 500
Possible cause | Suggested remedy |
---|---|
The outlet filter is closed off or blocked. |
Make sure that the outlet filter is not closed off or blocked. Try opening the spare (middle) filter to depressurize the Cellbag. You can connect this filter to the Exhaust pump and continue operations. If foam is clogging the outlet filter, reduce the rocking speed or the rock angle. (The Cellbag should be inflated so that it requires force to squeeze it 2 inches H2O ± 0.2 inches for WAVE Bioreactor 200 and 3 inches H2O ± 0.2 inches for WAVE Bioreactor 500/1000). However, it should not be so pressurized that creases form near the attachment points. The inlet airflow will shut off automatically (PV=0) if the pressure exceeds pressure setpoint +1 inch H2O gauge)

Make sure that the correct Exhaust pump is connected to the respective outlet filter. Check to see that the Exhaust pump is running.

Poorly inflated Cellbag |
Check that the Cellbag is rigidly inflated. A poorly inflated Cellbag will foam rapidly.
(Some amount of foam is typical. However, foam should not cover more than 50% of the surface area.) |
Too high rocking rate |
In many cases foam will subside after a few hours of operation. If foam is still excessive, reduce the rocking rate. Check that this rocking rate still provides a sufficient dissolved oxygen concentration.

Too big rock angle |
Reduce the rock angle. Reducing the angle will reduce foam while being able to maintain a higher rock rate. The WAVE Bioreactor base units are factory set to ±5° from the horizontal. This angle has been found to be optimal for most cell lines.
