Understanding extractables and leachables

Assessment of process equipment-related leachables (PERLs) that can impact drug product quality or patient safety is a critical regulatory requirement to be included in an overall validation strategy. Starting with compiling standardized component extractables data aligned to industry standards (see below), to assessing how these align with your process risk, and whether based on evaluation thresholds any additional mitigation, such as simulation or leachables testing are warranted, Cytiva can support your comprehensive risk evaluation, testing and validation requirements.

Compile available data

Standardized extractables data aligned to USP <665> and BioPhorum is increasingly available for many single-use components and filters (see table below).

Assess process risk

Check whether the available standardized data align with your process risk and conditions.

Evaluate thresholds

Determine if worst-case compound levels exceed risk evaluation limits (e.g., analytical evaluation threshold).

Mitigate risk

Use Cytiva Validation Services for model-solvent simulations or leachables studies that match your process, with toxicologic assessment where needed.

Extractables reports are available through two portals (see the far-right column in the summary table). To speed up your search, note the report identifier before accessing the portal.
